Selinexor plus ruxolitinib in JAK inhibitor–naïve myelofibrosis: Phase 3 SENTRY trial

Rapid readout: PEAK trial shows besiclastinib plus sunitinib boosts second-line GIST PFS and response, with manageable safety.

The Phase SENTRY trial evaluated the combination of selinexor and ruxolitinib compared to ruxolitinib alone in patients with myelofibrosis who had not previously taken a Janus kinase inhibitor. The combination therapy achieved rapid, deep, and sustained spleen volume reductions, alongside higher rates of variant allele frequency reduction and meaningful overall survival benefit. Both treatment arms demonstrated similar improvements in symptom reduction, while the combination maintained a manageable safety profile consistent with the individual agents.
