Darovasertib Plus Crizotinib vs Investigator’s Choice as First-Line Treatment for Patients with HLA-A2 Negative Metastatic Uveal Melanoma: Primary Results from the OptimUM-02 Trial

In this OncLive Rapid Readout, Marlana Orloff, MD, discusses primary results from the phase 2b portion of the OptimUM-02 trial evaluating darovasertib plus crizotinib versus investigator’s choice as first-line therapy for patients with HLA-A2–negative metastatic uveal melanoma. Orloff reviews the rationale for targeting PKC and MET in uveal melanoma, a disease with limited systemic treatment options and historically poor outcomes after metastasis. She details that darovasertib plus crizotinib produced a clinically meaningful and statistically significant progression-free survival benefit compared with investigator’s choice, along with higher objective response and disease control rates by blinded independent central review. The presentation also addresses safety findings, including treatment-related adverse events consistent with prior experience and relatively low rates of treatment-related serious adverse events and discontinuations. Although overall survival data remain immature, Orloff explains that the findings support darovasertib plus crizotinib as a potential new therapeutic standard.
