
Dr Prince on the Design of the Comparison of MAVORIC and Real-World Vorinostat

Miles Prince, MD, MBBS, an adjunct professor in the School of translational medicine at Monash University, professor and director of Cancer Immunology and Molecular Oncology at Epworth Healthcare, and a hematologist at the Peter MacCallum Cancer Center, discussed results for a comparison of data from the mogamulizumab-kpkc (Potelieo) arm in the phase 3 MAVORIC study (NCT01728805) to a real-world cohort of patients receiving vorinostat (Zolinza) which were presented at the
Prince began by noting that the cohort of real-world patients in the comparison who received vorinostat had also never received prior mogamulizumab—a notable characteristic since patients in the vorinostat arm of MARVORIC were permitted to cross over to receive mogamulizumab. Patients in each group of the comparison were matched via key criteria which was critical in allowing mogamulizumab in MAVORIC to be compared with real-world vorinostat, he said.
Prince then pointed out that before comparing overall survival (OS), investigators of the analysis had to first ensure that each group of patients were comparative to one another. In addition to each group being comparative to one another, Prince noted that the at similar follow-up, OS data for patients in the mogamulizumab arm was significantly higher than those who received real-world vorinostat. Moreover, Prince pointed out that the OS for the MAVORIC arm of the analysis was not reached after follow-up, which exceeded 3 years. Notably, the median OS of patients in the real-world vorinostat arm was 30.95 years. Prince noted that the difference between the 2 groups was statistically significant.
