WU-KONG28 Results for First-Line Sunvozertinib

Lyudmila Bazhenova, MD, of UC San Diego Moores Cancer Center, reviews the phase 3 WU-KONG28 trial of first-line sunvozertinib vs carboplatin plus pemetrexed in untreated EGFR exon 20 insertion–mutated non–small cell lung cancer.

Lyudmila Bazhenova, MD, of UC San Diego Moores Cancer Center, reviews the phase 3 WU-KONG28 trial of first-line sunvozertinib vs carboplatin plus pemetrexed in untreated EGFR exon 20 insertion–mutated non–small cell lung cancer. Median progression-free survival was 10.3 vs 7.5 months, the response rate was 59% vs about 30%, and immature median overall survival was 29.8 vs 28.8 months, with 90% of patients in the chemotherapy arm crossing over to sunvozertinib. Enrollment spanned 15 countries on 4 continents, and about two-thirds of patients were Asian. Pasi A. Jänne, MD, PhD, of Dana-Farber Cancer Institute, says many clinicians know sunvozertinib from later-line use, where it is US approved but hard to obtain. He does not think the Asian majority weakens the findings, since these cancers respond similarly to targeted therapy across ancestries. He calls an oral agent alone vs chemotherapy a higher bar and attributes the similar survival to crossover.
