ESMO 2021: Updates in Triple Negative Breast Cancer


Drs Joyce O’Shaughnessy, Sara Tolaney, Sara Hurvitz, and Mark Pegram examine recent trial data from ESMO 2021 on triple-negative breast cancer.

Data from the following clinical trials are discussed:

  • Durvalumab improves long-term outcome in TNBC: results from the phase 2 randomized GeparNUEVO study investigating neoadjuvant durvalumab in addition to an anthracycline/taxane based neoadjuvant chemotherapy in early triple-negative breast cancer (TNBC)
  • KEYNOTE-522: Phase 3 study of neoadjuvant pembrolizumab + chemotherapy vs placebo + chemotherapy, followed by adjuvant pembrolizumab vs placebo for early-stage TNBC - ESMO Plenary July 2021
  • 1190 - Event-free survival (EFS), overall survival (OS), and safety of adding veliparib (V) plus carboplatin (Cb) or carboplatin alone to neoadjuvant chemotherapy in triple-negative breast cancer (TNBC) after ≥4 years of follow-up: BrighTNess, a randomized phase 3 trial
  • LBA16 - KEYNOTE-355: Final results from a randomized, double-blind phase 3 study of first-line pembrolizumab + chemotherapy vs placebo + chemotherapy for metastatic TNBC
  • 258P - Analysis of patients (pts) without an initial triple-negative breast cancer (TNBC) diagnosis (Dx) in the phase 3 ASCENT study of sacituzumab govitecan (SG) in brain metastases-negative (BMNeg) metastatic TNBC (mTNBC)
